Effect of Di-2-ethylhexyl Phthalate on Renal Cytochrome P450 Monooxygenases in Surgically Evoked Subchronic Renal Failure of Rats

Abstract:
The effects of di-2-ethylhexyl phthalate (DEHP) were studied on the remnant kidneys of albino rats during subchronic renal failure (SCRF) induced by subtotal nephrectomy. Serum creatinine and urea increased significantly already on the seventh day of DEHP treatment compared with control groups. The amount of creatinine was 3.23 mg/dL and that of urea 100.8 mg/dL (457% and 240% increment, respectively) on the 21st day of investigation. Changes in the quantity of microsomal total cytochrome P450 and c ytochrome b5 were also determined, and histological studies undertaken. Both the renal microsomal P450 and b5 content increased in the remnant kidneys following the exposure to DEHP, suggesting an induction of enzymes. The elevation of cytochrome P450 level preceded that of the b5 and reached a higher extent at all times during the investigation. Microanatomically, the cortex showed hypertrophy, but the medulla was little affected. A great hypertrophy of glomeruli was observed. Proximal tubular epithelium showed intracellular vacuolization and degenerative changes.
